      Antioxidant Metabolism and Chlorophyll Fluorescence during the Acclimatisation to Ex Vitro Conditions of Micropropagated Stevia rebaudiana Bertoni Plants

          Abstract

          In this study, the functioning of antioxidant metabolism and photosynthesis efficiency during the acclimatisation of Stevia rebaudiana plants to ex vitro conditions was determined. A high percentage of acclimatised plants (93.3%) was obtained after four weeks. According to the extent of lipid peroxidation, an oxidative stress occurred during the first hours of acclimatisation. A lower activity of monodehydroascorbate reductase (MDHAR) than dehydroascorbate reductase (DHAR) was observed after 2 days of acclimatisation. However, after 7 days of acclimatisation, stevia plants activated the MDHAR route to recycle ascorbate, which is much more efficient energetically than the DHAR route. Superoxide dismutase and catalase activities showed a peak of activity after 7 days of acclimatisation, suggesting a protection against reactive oxygen species. Peroxidase activity increased about 2-fold after 2 days of acclimatisation and remained high until day 14, probably linked to the cell wall stiffening and the lignification processes. In addition, a progressive increase in the photochemical quenching parameters and the electronic transport rate was observed, coupled with a decrease in the non-photochemical quenching parameters, which indicate a progressive photosynthetic efficiency during this process. Taken together, antioxidant enzymes, lipid peroxidation, and chlorophyll fluorescence are proven as suitable tools for the physiological state evaluation of micropropagated plants during acclimatisation to ex vitro conditions.

            Steviol glycosides: chemical diversity, metabolism, and function.

            Steviol glycosides are a group of highly sweet diterpene glycosides discovered in only a few plant species, most notably the Paraguayan shrub Stevia rebaudiana. During the past few decades, the nutritional and pharmacological benefits of these secondary metabolites have become increasingly apparent. While these properties are now widely recognized, many aspects related to their in vivo biochemistry and metabolism and their relationship to the overall plant physiology of S. rebaudiana are not yet understood. Furthermore, the large size of the steviol glycoside pool commonly found within S. rebaudiana leaves implies a significant metabolic investment and poses questions regarding the benefits S. rebaudiana might gain from their accumulation. The current review intends to thoroughly discuss the available knowledge on these issues.

              Interaction between hydrogen peroxide and plant hormones during germination and the early growth of pea seedlings.

              Hydrogen peroxide (H(2)O(2)) increased the germination percentage of pea seeds, as well as the growth of seedlings in a concentration-dependent manner. The effect of H(2)O(2) on seedling growth was removed by incubation with 10 microm ABA. The H(2)O(2)-pretreatment produced an increase in ascorbate peroxidase (APX), peroxidase (POX) and ascorbate oxidase (AAO). The increases in these ascorbate-oxidizing enzymes correlated with the increase in the growth of the pea seedlings as well as with the decrease in the redox state of ascorbate. Moreover, the increase in APX activity was due to increases in the transcript levels of cytosolic and stromal APX (cytAPX, stAPX). The proteomic analysis showed that H(2)O(2) induced proteins related to plant signalling and development, cell elongation and division, and cell cycle control. A strong correlation between the effect of H(2)O(2) on plant growth and the decreases in ABA and zeatin riboside (ZR) was observed. The results suggest an interaction among the redox state and plant hormones, orchestrated by H(2)O(2), in the induction of proteins related to plant signalling and development during the early growth of pea seedlings.
