          Abstract

          The objective of this study was to use chlamydospores of the fungusPochonia chlamydosporia (isolates VC1 and VC4) against Toxocara canis eggs in a 15-day in vitro assay. One thousand T. canis eggs were placed in Petri dishes containing 2% water agar medium with different concentrations of chlamydospores (1,000, 10,000 or 100,000) of each fungal isolate of P. chlamydosporia (treated groups) and 1,000 eggs in Petri dishes without fungus (control group). Egg counts were performed to determine the ovicidal activity, which was classified as three effect levels: type 1, type 2 and type 3. Significant differences (P < 0.01) in egg destruction were found in comparison with the control group. The highest percentage of egg destruction was found in plates containing 100,000 chlamydospores (68.5% for VC1 and 70.5% for VC4). Chlamydospores of P. chlamydosporiawere effective in destroying T. canis eggs and may contribute in the future towards combating the eggs of this parasite.

          O objetivo do trabalho foi utilizar clamidósporos do fungoPochonia chlamydosporia (isolados VC1 e VC4) na destruição de ovos de Toxocara canis, num ensaioin vitro, realizado no intervalo de 15 dias. Em cada placa de Petri com ágar-água 2% foram vertidos 1.000 ovos deT. canis em 1.000, 10.000 ou 100.000 clamidósporos de cada isolado do fungo (grupos tratados). Foram realizadas as contagens para verificar a atividade ovicida, classificada em três níveis de efeito: tipo 1, tipo 2 e tipo 3. Os resultados demonstraram que houve diferença significativa (P < 0,01) na destruição dos ovos em relação aos ovos observados nas placas do grupo controle. O maior percentual de ovos destruídos foi observado nas placas contendo 100.000 clamidósporos (68,5% para VC1 e 70,5% para VC4). Clamidósporos do fungo P. chlamydosporia foram efetivos na destruição dos ovos de T. canis podendo contribuir no futuro para o combate aos ovos deste parasito.

          Application of in vitro anthelmintic sensitivity assays to canine parasitology: detecting resistance to pyrantel in Ancylostoma caninum.

          Resistance of the canine hookworm Ancylostoma caninum to anthelmintic therapy with pyrantel is an emerging problem in canine veterinary practice. Detecting anthelmintic resistance in parasites of pets is problematic because traditional resistance-monitoring techniques used with livestock parasites, such as the faecal egg count reduction test, are often impractical for use in small animals. We used two field-collected isolates of A. caninum in an abbreviated critical trial to test their pyrantel resistance status. The strains showed high-level and low-level resistance, with in vivo pyrantel efficacies of 28% and 71%, respectively. We noted a distinct worm density dependence effect on faecal egg count during the critical trial; egg counts in the dogs containing the low-level resistant isolate were 41% higher 6 days after drug treatment, despite the removal of 71% of the adult worms by the drug treatment. We then assessed four candidate in vitro assays for their ability to detect pyrantel resistance in A. caninum larvae, using these two isolates. The assays included a new format termed the larval arrested morphology assay (LAMA), based on observation of the effects of pyrantel on the body shape adopted by infective stage A. caninum larvae in vitro. Our data suggests that three of these assays, the LAMA, the larval motility assay (LMA), and larval feeding inhibition assay (LFIA) show promise with regards to detection of pyrantel resistance in A. caninum, but the complexity of the LFIA would likely limit its suitability for field studies. In vivo pyrantel efficacies of 28% and 71% in the two A. caninum isolates were associated with a 17-fold shift in the in vitro IC(50) values measured using the LAMA. Further testing with isolates of varying degrees of resistance is required to determine which of these assays is suitable as a rapid in vitro laboratory test for pyrantel resistance in A. caninum. The present study also indicates that potential exists for the novel LAMA or the LMA to be of use in detecting pyrantel resistance in the human hookworms, Necator americanus and Ancylostoma duodenale.

            Ocorrência de protozoários e helmintos em amostras de fezes de cães e gatos da cidade de São Paulo

            Foram examinadas, pelo Laboratório de Doenças Parasitárias do Departamento de Medicina Veterinária Preventiva e Saúde Animal, da Faculdade de Medicina Veterinária e Zootecnia da Universidade de São Paulo, no período de janeiro de 1991 a janeiro de 1995, amostras de 353 cães e de 187 gatos, com idades variadas e de diferentes áreas da cidade de São Paulo. Os métodos de diagnóstico empregados foram: centrífugo-flutuação em solução de sacarose e centrífugo-sedimentação em água-éter. Aproximadamente 45,0% das amostras de cães examinadas (160 amostras) apresentaram-se positivas e o percentual de ocorrência de parasitos, em relação ao total de amostras foi: Ancylostoma spp. 20,40; Toxocara canis 8,49; Giardia sp. 7,65; Cryptosporidium parvum 2,83; Cystoisospora spp. 2,55; Sarcocystis spp. 1,70; Hammondia heydorni 0,85; e Spirocerca lupi, Trichuris vulpis e Dipylidium caninum 0,28. Das 187 amostras recebidas, provenientes de gatos, somente oito eram negativas e observou-se a ocorrência (%) de Cystoisospora spp. 38,5; Toxocara sp. 34,22; Giardia sp. 16,04; Cryptosporidium parvum 14,44; Ancylostoma spp. 13,37; Dipylidium caninum 10,69; Sarcocystis spp. 8,56; Physaloptera spp. 4,81; oocistos tipo "Hammondia-Toxoplasma" 1,60; Strongyloides stercoralis 1,60; e Platynosomum concinum 1,07. Em cães, as associações mais freqüentemente observadas foram: Ancylostoma spp. + T. canis em oito amostras e Ancylostoma spp. + Giardia sp. e Ancylostoma spp. + T. vulpis em cinco amostras. Em gatos, Toxocara sp. e Cystoisospora spp., encontrados em 24 amostras, e Toxocara sp. + Ancylostoma spp. em cinco amostras, foram as associações mais comumente observadas.

              Biological control of helminths.

              As a potential component in future integrated parasite-control strategies, biological control by means of predacious fungi seems to be moving from a promising possibility toward becoming a reality, and the netforming nematode-destroying fungus Duddingtonia flagrans appears to be the candidate of choice. Not only has this fungus been found in, and isolated from, fresh sheep, cattle and horse faeces, but it also appears to be the only fungus that is able to consistently and significantly reduce the number of infective trichostrongyle larvae in faeces from animals fed fungal spores. Results from the last few years have shown that D. flagrans is able to trap and destroy free-living stages of the most important and common trichostrongylid larvae with very similar external life-cycles, as well as larvae of parasites with a slightly different transmission biology (Nematodirus spp., Dictyocaulus viviparus). The introduction of microfungi for biological control could be as part of a feed supplement or incorporated in feed-blocks presented to animals which are raised under relatively intensive conditions and constant surveillance. Apart from the special niche for organic farmers, such a product would be suited for horses, small ruminants (as either milking herds or housed daily for other reasons), cattle in special situations and free-roaming pigs. The most important constraint, still, for a major breakthrough in biological control in the latter is the lack of good antagonists against the long-lived and rather resistant infective stages of parasites, being transmitted as larvae inside the egg. Since the first Conference on Novel Approaches to the Control of Helminth Parasites of Livestock in Armidale, Australia, 1995, there has been a steady evolution within the area of biological control of parasitic nematodes. Today this principle is being exploited and tested out in almost all parts of the world, under various climatic conditions and production systems. Where, in the past, a large part of the work focused on cattle and to a lesser degree horse and sheep parasites, the focus of the research in many of the newly involved countries is on small ruminants, because of their importance to primarily small-scale farmers in local communities. Today research and trials are either on-going or being planned in many developing countries, as well as in countries in transition. The involvement of multinational agencies in addition to national and industrial interests is very welcome and should increase the chances and keep up the momentum for development and implementation of biological control in future animal production around the world.
