      Fabrication of gold/graphene nanostructures modified ITO electrode as highly sensitive electrochemical detection of Aflatoxin B1

          Abstract

          Aflatoxins (AFs) are a family of fungal toxins that produced in food and feed by two Aspergillus species (Aspergillus flavus and Aspergillus parasiticus). Several techniques have been reported for AFs detection including high-pressure liquid chromatography, enzyme-linked immunosorbent assay, surface plasmon resonance and recombinant immune blotting assay. But, these methods are disadvantaged because they consumed a long time for analysis; in addition, they required a piece of complicated and expensive equipment. Therefore, developing of inexpensive sensors with high selectivity and sensitivity for detecting of AFs levels without extensive sample preparation has received great attention. Several electrochemical AFs sensors have been reported; however, there is still a need for developing a new, simple and rapid electrochemical AFs sensor. Here, we have developed a new AFs sensor based on Au nanostructures/graphene nanosheets modified ITO substrate that could enhance the Raman effect and the electrochemical conductivity. The modified electrode was prepared based on layer-by-layer electrochemical deposition method. AFs antibody was immobilized onto the Au nanostructures/graphene nanosheets; then it was used as a probe for rapid, simple and cheap detection of AFs level using Raman spectroscopy and electrochemical techniques. Our results demonstrated that the developed system showed a simple, easy and sensitive sensor for monitoring low concentrations of AFB1 with a detection limit of about 6.9 pg/mL, also it allowed the determination of AFB1 in spiked food samples.

            An aptamer-based dipstick assay for the rapid and simple detection of aflatoxin B1.

            A rapid and simple dipstick assay based on an aptamer has been developed for the determination of aflatoxin B1 (AFB1). The dipstick assay format was based on a competitive reaction of the biotin-modified aptamer specific to AFB1 between target and cy5-modified DNA probes. Streptavidin and anti-cy5 antibody as capture reagents were immobilized at test and control lines on a membrane of the dipstick assay. After optimization, the limit of detection for the dipstick assay was 0.1 ng/ml AFB1 in buffer. The method was confirmed to be specific to AFB1, and the entire process of the assay can be completed within 30 min. Aqueous methanol (20%) provided a good extraction efficiency, and the matrix influence from corn extracts was successfully reduced through 2-fold dilution. The results of AFB1 analysis for corn samples spiked with known concentration of AFB1 by the dipstick assay and ELISA showed good agreement. The cut-off value of the dipstick assay for corn samples was 0.3 ng/g AFB1. Therefore, the dipstick assay is first reported and considered as a rapid, simple, on-site and inexpensive screening tool for AFB1 determination in grains as well as a corn.

              A simple and rapid optical biosensor for detection of aflatoxin B1 based on competitive dispersion of gold nanorods.

              This report illustrates a promising one-step and label-free optical biosensor for determination of aflatoxin B1 (AFB1) that is most commonly found in foods and highly dangerous even at very low concentrations. In this research, gold nanorods (GNRs) were employed as a sensing platform, which showed high stability under high ionic strength conditions without addition of any stabilizing agent. GNR-AFB1-BSA (bovine serum albumin) conjugates aggregated after mixing with free antibodies, resulting in significant changes in absorption intensity. At the same time the existence of AFB1 molecules in samples caused dispersion of nanorods, as a result of competitive immune-reaction with antibodies. By taking advantages of the competitive dispersion of GNRs, the developed method could effectively reduce false results caused by undesirable aggregation, which is a big problem for spherical gold nanoparticles. Absorption intensity of UV-vis spectra served as the sensing indicator, with dynamic light scattering (DLS) measurement as another sensing tool. The designed biosensing system could detect AFB1 in a linear range from 0.5 to 20ngmL(-1), with a good correlation coefficient of 0.99. And the limit of detection (LOD) was 0.16ngmL(-1), indicating an excellent sensitivity with absorbance result. The recoveries of the spiked AFB1 in real peanut samples ranged from 94.2% to 117.3%. Therefore the proposed nano-biosensor was demonstrated to be sensitive, selective, and simple, providing a viable alternative for rapid screening of toxins in agriculture products and foods.
