          Abstract

          An experimental oil-adjuvanted, inactivated vaccine against bovine herpesvirus type 1 (BHV-1.1), was produced and evaluated in its capacity to induce neutralizing antibodies against bovine herpesvirus type 1 (BHV-1, subtypes 1.1 and 1.2) and bovine herpesvirus type 5 (BHV-5). Cattle were vaccinated and revaccinated 90 days later. Antibodies were measured at days 0, 30, 90, 120, 180, 270 and 450 days after the first dose of vaccine (DPV). Antibody titres to BHV-1.1 and BHV-1.2 were significantly higher than to BHV-5 throughout the experiment. While all calves seroconverted to BHV-1.1 and BHV-1.2 after the first dose of vaccine, only two out of 23 (8,7%) calves seroconverted to BHV-5. However, after the booster injection all animals seroconverted to the three virus types. At 450 DPV, 79% (15/19 cattle) and 84% (16/19) were still positive for antibodies to BHV 1.1 and BHV 1.2, whereas 50% (10/19) of the calves remained seropositive for BHV-5. It was concluded that although a potent BHV-1 vaccine may induce crossreactive neutralizing antibodies to BHV-5, the levels of such antibodies are significantly lower and of shorter duration than antibodies to BHV-1.1 or BHV-1.2.

          Foi avaliada a capacidade de uma vacina experimental oleosa inativada contra o herpesvírus bovino tipo 1 induzir anticorpos contra o herpesvírus bovino tipo 1 (BHV-1.1 e BHV-1.2) e herpesvírus bovino tipo 5 (BHV-5). Bovinos foram vacinados com duas doses de vacina, aplicadas com um intervalo de 90 dias. Foi medido o título de anticorpos nos dias 0, 30, 90, 120, 180, 270 e 450 após a primeira vacinação (DPV). Os títulos de anticorpos contra BHV-1.1 e BHV-1.2 foram significativamente maiores que os contra BHV-5, ao longo do experimento. Enquanto todos os animais soroconverteram para BHV-1.1 e BHV-1.2 após a primeira dose de vacina, apenas dois em 23 animais (8,7%) soroconverteram para o BHV-5. No entanto, após o reforço, todos os animais soroconverteram contra BHV-1.1, 1.2 e BHV-5. Aos 450 DPV, 79% (15/19 animais) e 84% (10/19) ainda se encontravam soropositivos para BHV-1.1 e BHV-1.2, enquanto apenas 50% dos animais (10/19) se demonstraram positivos contra o BHV-5. Foi concluído que, embora uma vacina com alta capacidade de indução de anticorpos contra o BHV-1 possa induzir anticorpos capazes de neutralizar o BHV-5, os níveis de tais anticorpos são significativamente mais baixos e de menor duração do que aqueles induzidos contra BHV-1.1 e BHV-1.2.

          Meningoencefalite em bovinos causada por herpesvírus bovino-5 no Mato Grosso do Sul e São Paulo

          Quinze focos de meningoencefalite por herpesvírus bovino-5 (BHV-5) foram diagnosticados entre agosto de 1993 e dezembro de 1996, sendo 14 provenientes do estado do Mato Grosso do Sul e um do estado de São Paulo. A doença ocorreu em diversos municípios e em diferentes épocas do ano. Foram afetados bovinos de 6 a 60 meses de idade, com uma morbidade de 0,05% a 5% e letalidade próxima a 100%. Os sinais clínicos foram exclusivamente nervosos e o curso da enfermidade variou de 1 a 15 dias. As principais lesões histológicas detectadas foram meningite e encefalite difusa com malacia do córtex cerebral e presença de corpúsculos de inclusão intranucleares em astrócitos e neurônios. O vírus foi isolado do cérebro de 11 de um total de 12 animais, e sua identidade confirmada por imunoperoxidase, utilizando-se anticorpos monoclonais específicos. Os surtos de encefalite por BHV-5 representam 5% dos diagnósticos realizados em bovinos pelo Hospital Veterinário da Universidade Federal do Mato Grosso do Sul. Os resultados deste trabalho evidenciam a importância da doença no Mato Grosso do Sul e indicam a necessidade de incluir a encefalite por BHV-5 no diagnóstico diferencial de outras doenças do sistema nervoso de bovinos frequentes no Estado.

            Comparative studies of BHV-1 variants by in vivo--in vitro tests.

            The new encephalitogenic BHV-1.3 and previously characterized BHV-1 strains were studied with reference to their immunogenic and protective potency and their antigenic relationships using "in vitro" and "in vivo" tests. The "in vitro" results obtained by neutralization kinetics showed that the Los Angeles (LA) strain (BHV-1.1) and a vaginal isolate L-114 strain (BHV-1.2) had antigenic similarities. Conversely, the behavior of the encephalitogenic strain A-663 (BHV-1.3), was significantly distinct. The "in vivo" protection test was carried out in calves using LA and A-663 strains. Post-vaccination antibodies and challenge with A-663 strain showed that the immunogenic behavior and protective capacities of both strains were similar. Neutralization kinetics differences between BHV-1.1 and BHV-1.3 did not alter the "in vivo" protection against BHV-1.3 challenge.

              Experimental infection of neonatal calves with neurovirulent bovine herpesvirus type 1.3.

              A type of bovine herpesvirus, BHV-1.3, causes encephalitis in calves, whereas BHV-1.1 causes respiratory disease. Three colostrum-deprived calves and two colostrum-fed calves were inoculated with BHV-1.3 by intranasal aerosolization. Two colostrum-deprived calves were inoculated with BHV-1.1 by intranasal aerosolization. BHV-1.3-inoculated calves demonstrated severe encephalitis with minimal respiratory lesions, and BHV-1.1-inoculated calves demonstrated severe respiratory lesions and no clinical signs of neurologic disease. Calves fed colostrum that contained virus neutralizing antibodies were protected against neurologic disease. Colostrum-fed BHV-1.3-inoculated calves did not develop disease although they did become infected; virus was shed in respiratory secretions for 10-13 days postinoculation, similar to infected colostrum-deprived calves. BHV-1.3 was reactivated from a latent state from one colostrum-fed calf after administration of dexamethasone 60 days postinoculation. Histopathologic examination of the three colostrum-deprived BHV-1.3-inoculated calves revealed severe lesions of encephalitis. One of the two BHV-1.1-inoculated calves had one focal lesion of encephalitis. Virus was isolated from brain tissue of colostrum-deprived BHV-1.3-inoculated calves and from one BHV-1.1-inoculated calf. Immunohistochemical staining for BHV-1 antigen was observed in neurons from the colostrum-deprived BHV-1.3-inoculated calves.
