          Abstract

          Amostras de saliva não estimulada de 50 crianças e de suas mães foram analisadas, objetivando-se determinar a prevalência das espécies do grupo mutans. Alíquotas de saliva, pura e após diluição decimal seriada, foram semeadas no meio SB20 modificado, com bastão angulado em L, e incubadas em microaerofilia durante 72 horas a 37ºC. Após a contagem do número de ufc do grupo mutans, colônias características foram transferidas para o meio tioglicolato, incubado a 37ºC - 24 horas, e submetidas à identificação bioquímica. Estreptococos do grupo mutans foram detectados na saliva de 47 (94,0%) crianças e de 50 (100,0%) mães. Verificou-se que 31 (62,0%) pares mãe/filho albergavam a(s) mesma(s) espécie(s), sendo que 15 (30,0%) crianças e 24 (48,0%) mães estavam multicolonizadas (S. mutans + S. sobrinus), com uma média de ufc/ml correspondente a 1,28 x 106 e 3,57 x 106, respectivamente.

          Translated abstract

          The aim of this study was to determine the prevalence of species of the mutans group in samples of non-stimulated saliva from 50 children and their mothers. Aliquots of pure saliva were submitted to seriate decimal dilution, plated with the aid of the L glass rod onto SB20 modified medium, and incubated in a candle jar for 72 hours at 37ºC. After the count of the cfu of the mutans group, the characteristic colonies were transferred to thioglycolatte medium, incubated for 24hours at 37ºC and submitted to a biochemical identification scheme. Streptococci of the mutans group were detected in the saliva of 47 (94.0%) children, and in 50 (100.0%) mothers. S. mutans and S. sobrinus were detected in 15 (30.0%) children, and in 24 (48.0%) mothers who were multicolonized, with the cfu number corresponding to 1.28 x 106 and 3.57 x 106, respectively. The same species of the mutans group were harbored by 31 (62.0%) mother-child pairs.

            Caries prevalence and salivary mutans streptococci in 0-2-year-old children of Japan.

            The prevalence of caries and the number and species distribution of salivary mutans streptococci in 356 children (aged 0-2 yr old) in Japan were examined twice at an interval of 1 yr. No mutans streptococci were isolated from the predentate children. The detection rate of mutans streptococci and the prevalence of caries increased with age. Mutans streptococci were isolated from 39.9% of the subjects. The majority of isolates were identified as Streptococcus mutans. The concentration of mutans streptococci correlated with the number of erupted teeth (r = 0.339). In addition, the concentration of mutans streptococci was more closely correlated to the caries prevalence of the next year (r = 0.465) than that of the year when the salivary concentration of mutans streptococci was first evaluated (r = 0.368). The children who had no caries but harbored mutans streptococci in the first year showed significantly higher caries prevalence in the next year than did the children with neither caries nor mutans streptococci. These results indicate that the establishment of mutans streptococci is associated with caries initiation in early childhood.

              Comparative recovery of Streptococcus mutans on five isolation media, including a new simple selective medium.

              For the isolation of Streptococcus mutans, several selective media have been developed, of which Mitis-Salivarius Sucrose Bacitracin agar (MSB) is the most widely used (Gold et al., 1973). Recently, the Trypticase Yeast-Extract Cystine agar medium (TYC, de Stoppelaar et al., 1967) was modified into a selective medium for S. mutans, called Trypticase Yeast-Extract Cystine Sucrose Bacitracin (TYCSB, van Palenstein Helderman et al., 1983). The aim of this study was to compare the recovery of S. mutans from clinical samples on Mitis-Salivarius agar (MS), MSB, TYC, and TYCSB. Further, a new simple selective medium for S. mutans was introduced. This medium, called TSY20B, was supposed to have the same qualities as TYCSB, but its preparation is less laborious. One hundred eighty-five plaque and saliva samples from 37 subjects were plated on MS, MSB, TYC, and TYCSB, and 285 samples from 23 subjects were plated on TYCSB and TSY20B. All plates were incubated at 37 degrees C in a 91% N2, 5% CO2, 4% H2 atmosphere for five days. The S. mutans counts on TYC and TYCSB were significantly higher than on MS or MSB by almost a factor of 10. Seventy-seven percent of the samples gave higher S. mutans counts on TYCSB than on MSB. Especially, samples with high S. mutans d/g numbers gave lower S. mutans counts on MSB. These data clearly indicate that MSB agar is inhibitory for S. mutans and should not be used. An additional advantage of TYCSB over MSB agar is the possibility of distinguishing S. mutans serotypes d/g from other serotypes.(ABSTRACT TRUNCATED AT 250 WORDS)
