      Utilisation of Mangifera indica plant extracts and parts in antimicrobial formulations and as a pharmaceutical excipient: a review

          Abstract

          Background

          Antimicrobial resistance and the environmental threat posed by some synthetic antimicrobial agents necessitate more research into development of novel pharmaceutical products that are environmentally friendly. Also, the use of plant derived excipients is growing and opening up new avenue to solve current drug delivery issues in the pharmaceutical industry.

          Main body

          This review summarizes studies related to the antimicrobial property of Mangifera indica extracts, possible mechanisms of antimicrobial action and antimicrobial formulations from the plant and overview of researches relating to the use of M. indica as a pharmaceutical excipient. Electronic searches were conducted on databases such as Pub Med, Wiley Online Library (WOL) and Google Scholar with focus on published articles relating to M. indica. Inclusion and exclusion criteria include publications relating to antimicrobial properties of M. indica extracts, its antimicrobial formulations and its use as a pharmaceutical excipient. The electronic searches yielded about 190 articles. From the studies reviewed, the mechanisms of action of phytochemicals described corroborate the antimicrobial activity exhibited by M. indica extracts and its selected formulations. In addition, mango pectin was observed to possess potential as a pharmaceutical excipient. Very few previous review articles based their focus on incorporating mechanism of action of phytochemicals with antimicrobial activity.

          This review examined antimicrobial properties of M. indica extracts and formulations, major phytochemicals in the plant parts and their possible modes of action. In addition, the study assessed the use of natural polymer derived from mango plant as excipients in pharmaceutical and pharmacological preparations.

          Conclusion

          The study concluded that effective antimicrobial activity of mango plant extracts and formulations requires synergy of actions among various phytochemical constituents of the extract or formulation. It is recommended that more researches focused on discovery of new phytochemicals in M. indica, their mechanisms of action and effective utilization of the plant in the pharmaceutical industry should be further explored.

          Flavonoids: an overview

          Flavonoids, a group of natural substances with variable phenolic structures, are found in fruits, vegetables, grains, bark, roots, stems, flowers, tea and wine. These natural products are well known for their beneficial effects on health and efforts are being made to isolate the ingredients so called flavonoids. Flavonoids are now considered as an indispensable component in a variety of nutraceutical, pharmaceutical, medicinal and cosmetic applications. This is attributed to their anti-oxidative, anti-inflammatory, anti-mutagenic and anti-carcinogenic properties coupled with their capacity to modulate key cellular enzyme function. Research on flavonoids received an added impulse with the discovery of the low cardiovascular mortality rate and also prevention of CHD. Information on the working mechanisms of flavonoids is still not understood properly. However, it has widely been known for centuries that derivatives of plant origin possess a broad spectrum of biological activity. Current trends of research and development activities on flavonoids relate to isolation, identification, characterisation and functions of flavonoids and finally their applications on health benefits. Molecular docking and knowledge of bioinformatics are also being used to predict potential applications and manufacturing by industry. In the present review, attempts have been made to discuss the current trends of research and development on flavonoids, working mechanisms of flavonoids, flavonoid functions and applications, prediction of flavonoids as potential drugs in preventing chronic diseases and future research directions.

            Flavonoid antioxidants: chemistry, metabolism and structure-activity relationships.

            Flavonoids are a class of secondary plant phenolics with significant antioxidant and chelating properties. In the human diet, they are most concentrated in fruits, vegetables, wines, teas and cocoa. Their cardioprotective effects stem from the ability to inhibit lipid peroxidation, chelate redox-active metals, and attenuate other processes involving reactive oxygen species. Flavonoids occur in foods primarily as glycosides and polymers that are degraded to variable extents in the digestive tract. Although metabolism of these compounds remains elusive, enteric absorption occurs sufficiently to reduce plasma indices of oxidant status. The propensity of a flavonoid to inhibit free-radical mediated events is governed by its chemical structure. Since these compounds are based on the flavan nucleus, the number, positions, and types of substitutions influence radical scavenging and chelating activity. The diversity and multiple mechanisms of flavonoid action, together with the numerous methods of initiation, detection and measurement of oxidative processes in vitro and in vivo offer plausible explanations for existing discrepancies in structure-activity relationships. Despite some inconsistent lines of evidence, several structure-activity relationships are well established in vitro. Multiple hydroxyl groups confer upon the molecule substantial antioxidant, chelating and prooxidant activity. Methoxy groups introduce unfavorable steric effects and increase lipophilicity and membrane partitioning. A double bond and carbonyl function in the heterocycle or polymerization of the nuclear structure increases activity by affording a more stable flavonoid radical through conjugation and electron delocalization. Further investigation of the metabolism of these phytochemicals is justified to extend structure-activity relationships (SAR) to preventive and therapeutic nutritional strategies.

              Flavonoids: a review of probable mechanisms of action and potential applications.

              The aim of this review, a summary of the putative biological actions of flavonoids, was to obtain a further understanding of the reported beneficial health effects of these substances. Flavonoids occur naturally in fruit, vegetables, and beverages such as tea and wine. Research in the field of flavonoids has increased since the discovery of the French paradox,ie, the low cardiovascular mortality rate observed in Mediterranean populations in association with red wine consumption and a high saturated fat intake. Several other potential beneficial properties of flavonoids have since been ascertained. We review the different groups of known flavonoids, the probable mechanisms by which they act, and the potential clinical applications of these fascinating natural substances.
