      Development of enzyme-linked immunosorbent assays based on recombinant MSP1a and MSP2 of Anaplasma marginale

          Abstract

          Indirect enzyme-linked immunosorbent assays (ELISAs) based on recombinant MSP1a and MSP2 from a Brazilian isolate of Anaplasma marginale were developed to detect antibodies against this rickettsia in cattle. The high sensitivities (99% for both tests) and specificities (100% for both tests) were confirmed with sera from cattle positive or negative for A. marginale antibodies, respectively, by immunofluorescent antibody test. By the analysis of 583 sera from cattle of three regions of the state of Pernambuco, Brazil, the agreement between both tests was high, with a kappa index of 0.89. The similar performances of the ELISAs suggest that both tests can be used in epidemiological surveys for detection of antibodies to A. marginale in cattle.

          Detection of antibodies in trypanosome-infected cattle by means of a microplate enzyme-linked immunosorbent assay.

          A micromodification of the enzyme-linked immunosorbent assay (ELISA) was evaluated for its potential application in the immunodiagonsis of bovine trypanosomiasis. Serum samples from infected and non-infected Zebu cattle and samples from Friesian cattle with experimental infections were exmained for the presence of trypanosomal antibodies. There were significant differences between the microELISA values obtained with samples from infected and non-infected cattle. During the course of infection microELISA values were found to fluctuate and the antibody response varied in individual animals. The test did not distinguish between infections with Trypanosoma brucei, T. vivax and T. congolense. There were no cross-reactions between trypanosome antigens and serum samples from cattle infected with T. theileri, Theileria parva, Th. mutans, Th. annulata, Babesia divergens and Anaplasma marginale.

            Molecular basis for surface antigen size polymorphisms and conservation of a neutralization-sensitive epitope in Anaplasma marginale.

            Anaplasmosis is one of several tick-borne diseases severely constraining cattle production and usage in many parts of the world. Cattle can be protected from anaplasmosis by immunization with major surface protein 1, a surface protein of Anaplasma marginale carrying a neutralization-sensitive epitope. Marked size polymorphisms exist among different isolates of A. marginale in the AmF105 subunit of major surface protein 1, yet all isolates still contain the neutralization-sensitive epitope. To clarify the basis for these observations, the mspl alpha gene encoding AmF105 was cloned from four isolates and sequenced. The encoded polypeptides share a high degree of overall homology between isolates but contain a domain with various numbers of tandemly repeated sequences and three regions of clustered amino acid substitutions outside the repeat domain. The polypeptide size differences are completely explained by the variations in the numbers of tandem repeat units. We have mapped the neutralization-sensitive epitope to a sequence that is present within each repeat unit. These results identify a basis for size polymorphisms of the surface polypeptide antigen concomitant with B-cell epitope conservation in rickettsiae.
