      Pseudorabies Virus Variant in Bartha-K61–Vaccinated Pigs, China, 2012

          Abstract

          The widely used pseudorabies virus (PRV) Bartha-K61 vaccine has played a key role in the eradication of PRV. Since late 2011, however, a disease characterized by neurologic symptoms and a high number of deaths among newborn piglets has occurred among Bartha-K61–vaccinated pigs on many farms in China. Clinical samples from pigs on 15 farms in 6 provinces were examined. The PRV gE gene was detectable by PCR in all samples, and sequence analysis of the gE gene showed that all isolates belonged to a relatively independent cluster and contained 2 amino acid insertions. A PRV (named HeN1) was isolated and caused transitional fever in pigs. In protection assays, Bartha-K61 vaccine provided 100% protection against lethal challenge with SC (a classical PRV) but only 50% protection against 4 challenges with strain HeN1. The findings suggest that Bartha-K61 vaccine does not provide effective protection against PRV HeN1 infection.

          Antiviral effect of diammonium glycyrrhizinate and lithium chloride on cell infection by pseudorabies herpesvirus

          Diammonium glycyrrhizin (DG), a salt from glycyrrhizinate (GL) that is a major active component of licorice root extract with various pharmacological activities was investigated for its inhibitory effect on pseudorabies virus (PrV) infection. In parallel, lithium chloride (LiCl), a chemical reagent with potential antiviral activity was compared with DG for their inhibitory ability against PrV infection in vitro. Virus plaque-reduction assays, PCR and RT-PCR analysis indicated that both drugs inhibited cell infection by PrV. Moreover, addition of the drugs resulted in fewer apoptotic cells during PrV infection.

            Genetic diversity and phylogenetic analysis of glycoprotein 5 of PRRSV isolates in mainland China from 1996 to 2006: coexistence of two NA-subgenotypes with great diversity.

            GP5, the most important neutralizing antigen of porcine reproductive and respiratory syndrome virus (PRRSV), has the highest genetic diversity among isolates. To more fully understand the extent of genetic diversity of PRRSV in China, we analyzed and compared the GP5 sequences of 42 PRRSV isolated from 1996 to 2006 in mainland China. We found that all of the Chinese isolates examined belong to the North American (NA) type. Among them two highly diverse subgroups were clearly demarcated on the NA-genotype phylogenetic tree. All the subgroup 1 isolates were found to be high variable in the primary neutralizing epitope and the viruses were geographically restricted to regions in southeast China. The subgroup 2 isolates shared a high identity with MLV vaccine and its parent virus VR-2332. These results may contribute to the knowledge of PRRSV epidemiology in China, and may help to explain the low efficiency of MLV or killed CH-1a vaccine to protect the subgroup 1 virus infected pigs, and the great genetic diversity should be taken into consideration for control and preventive measures.

              Eradication of Aujeszky's disease in Germany.

              Aujeszky's disease (AD) manifested itself in both German states in 1960. Owing to the historical development, in the subsequent two decades, the development of the disease and of its control in the Western and Eastern parts of Germany went different ways. This article describes differences and particularities in the development of AD in Germany leading to the establishment of a national AD eradication programme after re-unification of the two German states at the beginning of the last decade. The basic principles of the German AD eradication programme are described, and the results of 10 years of efforts to control the disease are presented and discussed. Without any doubt, as in other European countries, implementation of the national eradication programme resulted in a considerable progress in the eradication of AD. Since the eradication programme has been established in 1989, particularly in West Germany, the number of AD outbreaks has decreased steadily from about 2000 cases in 1987 to 0 cases recorded in 2001. Recently, Germany has been declared as officially AD-free by the European Commission.
